Glycosaminoglycans in the nonswelling corneal stroma of dogfish shark.

چکیده
Qualitative and quantitative determinations of glycosaminoglycans in the nonswelling corneal stroma of the spiny dogfish shark, Squalus acanthias, were made with the use of a cetylpyridinium chloride-cellulose microfractionation technique. Dogfish stroma was found to contain predominantly highly sulfated galactosaminoglycans forming approximately 75 per cent of the total glycosaminoglycans when calculated on the basis of hexosamine content. The remaining portion of corneal glycosaminoglycans was identified as glucosaminoglycans with a molar ratio of sulfate to hexosamine higher than one. Quantitative differences were observed between the anterior and posterior parts of the dogfish corneal stroma both as to the concentration of hexosamine and the ratio of galactosamine to glucosamine.

The relationship between morphology and transparency in the nonswelling corneal stroma of the shark.
The ultrastructure of the corneal stroma of the spiny dogfish, Squalus acanthias, was studied in order to determine whether there was a morphologic basis for the alleged inability of the elasmobranch cornea to swell, and for the possibly related maintenance of corneal transparency when it is immersed in distilled water. The most apparent morphologic explanation for the nonswelling properties wa...
متن کاملUltrastructure Organization of Collagen Fibrils and Proteoglycans of Stingray and Shark Corneal Stroma
We report here the ultrastructural organization of collagen fibrils (CF) and proteoglycans (PGs) of the corneal stroma of both the stingray and the shark. Three corneas from three stingrays and three corneas from three sharks were processed for electron microscopy. Tissues were embedded in TAAB 031 resin. The corneal stroma of both the stingray and shark consisted of parallel running lamellae o...
متن کاملElectrical parameters of the isolated cornea of the dogfish, Squalus acanthias.
The electrical potential difference and electrical resistance of the nonswelling cornea of the dogfish, Squalus acanthias, were examined. It was found that routine procedures used in the procurement of fish invariably produce damage to the corneal epithelium which affects electrical measurements and possibly composition of the aqueous humor. We found no electrical evidence of ionic pumps in the...
متن کاملStromal sodium binding after glycosaminoglycan digestion.
The glycosaminoglycans of isolated rabbit corneal stroma, clamped between two lucite plates at near normal hydration, were digested with testicular hyaluronidase in saline solution. After equilibration with 0.9 per cent saline solution alone the sodium and chloride content of the stroma was determined. Chloride was in equilibrium with both normal and hyaluronidase-treated stroma, allowing use o...
متن کاملThe biosynthesis in vitro of keratan sulphate in bovine cornea.
1. Bovine corneas were incubated in vitro with [U-(14)C]glucose. 2. The glycosaminoglycans of corneal stroma were isolated and fractionated on cetylpyridinium chloride-cellulose columns. The major components were keratan sulphate (71%), chondroitin 4-sulphate (17%) and chondroitin 6-sulphate (4%). 3. The acid-soluble nucleotides and intermediates of glycosaminoglycan biosynthesis of corneal str...
